Human TNNI1/Troponin I, slow skeletal muscle ELISA Kit

A high-quality sandwich ELISA from Assay Genie designed for the quantitative measurement of human TNNI1 (Troponin I, slow skeletal muscle). The kit uses a double-antibody sandwich format and reports a sensitivity of 28.125 pg/mL with a standard range of 46.875–3000 pg/mL. Detection is colorimetric and the kit is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. All components should be stored at 2–8 °C for up to 12 months.

Troponin I, slow skeletal muscle is a key protein that regulates muscle contraction, and abnormal levels have been linked to various muscle disorders and cardiac conditions, making it a valuable biomarker in research into muscle function and disease pathology. The workflow follows a standard sandwich ELISA protocol: equilibrate reagents and the pre-coated plate, incubate standards and samples at 37 °C for 90 minutes, add biotin-labelled detection antibody and incubate for 60 minutes at 37 °C, add HRP-streptavidin (SABC) for 30 minutes at 37 °C, develop with TMB substrate in the dark for 10–20 minutes, stop the reaction, and read absorbance at 450 nm. Reported linearity falls between 82% and 102% across serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions, with average spike-and-recovery values of 98% (serum), 97% (EDTA plasma) and 97% (heparin plasma).

Sample preparation guidance is provided for serum (allow blood to clot, centrifuge and collect supernatant), plasma (collect in anticoagulant tubes and centrifuge), tissue homogenate (homogenise in PBS with protease inhibitors and centrifuge), cell culture supernatant (centrifuge to clarify) and cell lysate (lyse with buffer containing protease inhibitors and collect the protein supernatant).
