TNFRSF9 (4-1BB/CD137) Immunoassay for Immune Checkpoint Research

TNFRSF9, also known as 4-1BB or CD137, is a costimulatory receptor of the tumor necrosis factor receptor superfamily. It is transiently upregulated on activated CD4+ and CD8+ T lymphocytes, regulatory T cells, natural killer (NK) cells, and dendritic cells. Engagement of 4-1BB by its trimeric ligand 4-1BBL (TNFSF9) drives T-cell proliferation, sustains cytotoxic effector function, enhances memory T-cell formation, and amplifies NK-cell activity. These properties have positioned TNFRSF9 as a focal target in cancer immunotherapy, including clinical-stage agonist antibodies such as urelumab and utomilumab, and in chimeric antigen receptor (CAR) T-cell engineering.

The Assay Genie Human TNFRSF9 PharmaGenie ELISA Kit is a quantitative sandwich-format immunoassay used to measure soluble human TNFRSF9 in cell culture supernatants, plasma samples, and serum samples. Each kit is supplied with a pre-coated microplate and complete reagents for 96 individual determinations in a single-plate workflow, supporting immunology, oncology, and translational biomarker laboratories that study costimulatory signaling and T-cell exhaustion.

Common research applications include tracking soluble 4-1BB shedding as a pharmacodynamic readout in immuno-oncology studies, profiling immune activation following vaccination, and characterizing autoimmune disease activity where elevated TNFRSF9 levels have been documented in patient cohorts. Colorimetric readout at the assay's recommended wavelength enables straightforward plate-reader integration for high-throughput screening campaigns.
