Description

This sandwich ELISA kit from Assay Genie enables quantitative detection of human TNFAIP3 (TNF alpha-induced protein 3, also known as A20) in serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. The kit uses a colorimetric, double-antibody sandwich format with a reported sensitivity of 0.188 ng/mL and a detection range of 0.313–20 ng/mL. Components are stored at 2-8°C for up to 12 months.

The workflow consists of six steps: reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-streptavidin incubation at 37°C for 30 minutes, TMB color development in the dark for 10–20 minutes, and stop reaction with absorbance read at 450 nm. Reported linearity (n=5) across 1:2, 1:4, and 1:8 dilutions was 82–104% for serum, 81–101% for EDTA plasma, and 86–105% for heparin plasma. Recovery averages were 99% for serum, 100% for EDTA plasma, and 97% for heparin plasma.

TNFAIP3 (A20) is described as a critical regulator of inflammatory and immune responses, controlling signaling pathways that mediate inflammation and cell survival. Its dysregulation has been linked to autoimmune diseases, inflammatory disorders, and cancer, making it a target of interest for therapeutic intervention research.
