Human TNF-beta ELISA Kit

A sandwich ELISA kit from Assay Genie designed for the quantitative measurement of TNF-beta (also known as Lymphotoxin alpha, LTA, TNFBeta or LT-alpha) in human samples. The assay uses a double-antibody sandwich format with colorimetric detection, offering a sensitivity of 9.375 pg/mL and a measurable range of 15.625–1000 pg/mL.

The kit is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. Sample preparation guidance includes allowing blood to clot and centrifuging serum, collecting plasma with anticoagulants such as EDTA or heparin, clarifying cell culture supernatant, and homogenising tissue in PBS with protease inhibitors.

A standard protocol involves equilibrating reagents and loading standards, samples and controls onto the pre-coated plate, incubating at 37 °C to allow antigen binding, adding a biotin-labelled detection antibody, then an HRP-Streptavidin conjugate, developing colour with TMB substrate in the dark, and reading absorbance at 450 nm after adding stop solution. Reagents should be stored at 2–8 °C. Linearity and recovery have been demonstrated across serum, EDTA plasma and heparin plasma at multiple dilutions.
