The Human TNF-alpha ELISpot Pair from Assay Genie supplies a matched set of capture and biotinylated detection antibodies for researchers building custom immunospot assays to count individual TNF-alpha secreting cells.

TNF-alpha is a central pro-inflammatory cytokine released by macrophages, T lymphocytes, and NK cells during innate and adaptive immune responses. Enumerating antigen-specific TNF-alpha producers at single-cell resolution is a common readout in vaccine development, infectious-disease studies, autoimmune research, and transplant immunology workflows.

As a "pair" format rather than a complete kit, the product contains only the antibody reagents, giving laboratories freedom to choose their preferred membrane plate (commonly PVDF-backed), cell density, and detection chemistry. A typical workflow seeds 100-200 µL of cell suspension per well, incubates overnight at 37 °C in a humidified CO2 incubator, then develops spots with a streptavidin-enzyme conjugate for analysis on an automated reader or stereomicroscope. Reported sensitivity reaches the single-cell level (detection typically below 1 pg/mL), with results expressed as spot-forming units (SFU) per million PBMCs.

The pair configuration is also convenient for multiplex-style work, since the same streptavidin-HRP detection reagent can be reused across additional cytokine pairs from the same donor sample. Capture antibody is supplied in a vial and biotinylated detection antibody in a separate vial, both intended for refrigerated storage. For research use only; not for diagnostic procedures.
