Description
A colorimetric sandwich ELISA from Assay Genie designed to measure human TMPRSS4 (Transmembrane Protease, Serine 4) levels in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a double-antibody sandwich format with biotin-labeled detection antibody, HRP-streptavidin (SABC), and TMB substrate read at 450 nm. Reported sensitivity is 0.469 ng/mL and the assay range is 0.781–50 ng/mL. Reagents are stored at 2–8°C for 12 months.
Linearity across serum, EDTA plasma, and heparin plasma (n = 5) at 1:2, 1:4, and 1:8 dilutions ranged from 81% to 105%. Spike-and-recovery performance is reported for serum (range 90–104%, average 95%), EDTA plasma (range 85–104%, average 96%), and heparin plasma (range 86–98%, average 92%). The protocol involves equilibrating reagents, a 90-minute primary incubation at 37°C, a 60-minute biotin-antibody incubation, a 30-minute SABC incubation, and 10–20 minutes of TMB color development in the dark before stop solution is added.
TMPRSS4 is described as a protein involved in cellular processes including cell signaling, proliferation, and migration, with dysregulation linked to cancer progression and metastasis, making it relevant for cancer biology research. Synonyms listed include TMPRSS4, Transmembrane protease serine 4, Channel-activating protease 2, CAPH2, Membrane-type serine protease 2, and MT-SP2 (UniProt Q9NRS4).
