Human TLR3/Toll-like receptor 3 ELISA Kit

Assay Genie provides a sandwich ELISA kit for the precise measurement of Toll-Like Receptor 3 (TLR3, also known as CD283) in human samples. The kit is positioned for research into innate immunity, where TLR3 recognises viral double-stranded RNA and initiates immune responses, as well as for studying conditions linked to TLR3 dysregulation, including inflammatory and autoimmune diseases.

Validated for serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples, the assay is run in a double-antibody sandwich format. Linearity has been characterised in serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions, and recovery averages of 95% (serum), 99% (EDTA plasma) and 98% (heparin plasma) are reported.

The protocol begins by equilibrating reagents to room temperature and loading standards, samples and controls onto the pre-coated plate, followed by incubation at 37°C for 90 minutes. A biotin-labelled detection antibody is then added (60 minutes at 37°C), followed by HRP-Streptavidin (30 minutes at 37°C), TMB substrate (10–20 minutes in the dark), stop solution, and absorbance is read at 450 nm. Supplied components include a dismountable microplate, lyophilised standard, biotin-labelled antibody concentrate, HRP-Streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25× wash buffer and plate sealer.
