Description
Designed for quantitative measurement of TIRAP (Toll Interleukin-1 Receptor Domain Containing Adaptor Protein) in human samples, this Sandwich ELISA Kit from Assay Genie targets an adaptor protein that plays a key role in the Toll-like receptor signalling pathway, which is crucial for the activation of the immune response. Dysregulation of TIRAP has been associated with various inflammatory and autoimmune diseases, making it a significant biomarker for immunological research.
The double-antibody sandwich assay uses colorimetric detection with a sensitivity of 18.75 pg/mL and a range of 31.25–2000 pg/mL. It is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported linearity (n=5) at 1:2, 1:4, and 1:8 dilutions is 85–105% / 96–105% / 90–98% for serum, 83–98% / 86–97% / 83–100% for EDTA plasma, and 89–98% / 81–100% / 85–96% for heparin plasma. Recovery ranges are 86–103% (average 90%) for serum, 85–105% (average 96%) for EDTA plasma, and 86–103% (average 96%) for heparin plasma.
The procedure includes equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes, HRP-Streptavidin binding for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after addition of stop solution. The kit should be stored at 2-8°C for 12 months.
