Description

A colorimetric Sandwich ELISA from Assay Genie for the quantitative measurement of Thrombin-Antithrombin Complex (TAT) in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The TAT complex is a critical marker of coagulation and thrombosis, reflecting the balance between pro-coagulant thrombin and anti-coagulant antithrombin. Dysregulation of the complex has been implicated in sepsis, disseminated intravascular coagulation (DIC), and other thromboembolic disorders, making the kit a useful resource for studying these conditions and exploring therapeutic interventions.

Assay Genie reports a sensitivity of 46.875 pg/mL and a detection range of 78.125–5000 pg/mL using a double-antibody sandwich format. Linearity for EDTA plasma (n=5) was 85–99% at 1:2, 85–99% at 1:4, and 82–100% at 1:8, while heparin plasma (n=5) gave 83–100%, 81–95%, and 81–100% across the same dilutions. Recovery was 89–100% (average 97%) in EDTA plasma and 88–101% (average 95%) in heparin plasma. The six-step procedure involves equilibrating reagents and TMB substrate, incubating standards and samples at 37°C for 90 minutes, adding a biotin-labeled detection antibody for 60 minutes, incubating with HRP-Streptavidin (SABC) for 30 minutes, developing with TMB substrate for 10–20 minutes in the dark, and reading absorbance at 450 nm after stop solution is added. Reagents are stored at 2–8°C for 12 months.

Sample preparation guidance is provided for serum (allow blood to clot, centrifuge at 1000 × g for 20 minutes), plasma (collect using anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2–8°C), tissue homogenate (homogenize in PBS with protease inhibitors), cell culture supernatant (centrifuge at 2500 rpm for 5 minutes), and cell lysate (lyse with buffer containing protease inhibitors). Assay Genie's technical support team can advise on additional sample types such as body fluids.
