Description

Manufactured by Assay Genie, this Sandwich ELISA Kit is designed for the in vitro quantitative determination of Human Thioredoxin (TRX) protein concentrations in serum, plasma, and other biological fluids. It is intended for Research Use Only and detects the target using a colorimetric, double-antibody sandwich method. The kit has a reported sensitivity of 0.188 ng/ml and an assay range of 0.313–20 ng/ml, with a shelf-life of 6 months at 4 °C.

Reported performance characteristics include intra-assay CV <8% and inter-assay CV <10%, with recovery ranges from spiked matrices of 86–93% in serum (average 91%), 88–105% in EDTA plasma (average 93%) and 98–102% in UFH plasma (average 99%). Linearity across serial dilutions of spiked samples has been demonstrated in serum, EDTA plasma and UFH plasma. Thioredoxin, encoded by the TXN gene (aliases Trx, Trx1, TXN1) on chromosomal region 9q31 (UniProt P10599, NCBI Gene ID 7295), is a homodimeric, disulfide-linked protein of approximately 9,452 Da that participates in redox reactions via reversible oxidation of its active-center dithiol and contributes to cellular responses to nitric oxide and ionizing radiation. It is localised to the cytoplasm, cytosol, mitochondrion, nucleoplasm and nucleus.

The reagent set includes a dismountable ELISA microplate, lyophilized standard, sample/standard dilution buffer, biotin-labelled antibody, antibody dilution buffer, HRP-streptavidin conjugate (SABC), SABC dilution buffer, TMB substrate, stop solution, wash buffer and plate sealers. The accompanying protocol outlines a sandwich ELISA workflow involving standard and sample incubation at 37 °C, biotinylated detection antibody incubation, SABC addition, TMB colour development and absorbance reading at 450 nm. Detailed sample preparation guidance is provided for serum, plasma (EDTA or heparin), urine, cerebrospinal fluid, cell culture supernatant, cell lysates, tissue homogenates, tissue lysates and breast milk.
