Built around a sandwich immunoassay architecture, the kit pairs a pre-titered capture antibody immobilized on a 96-well microplate with a biotinylated secondary antibody and a streptavidin-horseradish peroxidase conjugate, producing luminescent output on a CLIA-compatible reader. The calibrator series is calibrated against recombinant full-length human TGFBI, and recombinant TGFBI spiked into serum typically recovers in the 85-115% range with intra-assay CV below 10%, supporting reliable quantitation of the endogenous protein. The secreted 68 kDa matricellular ligand contains four FAS1 fasciclin domains and mediates adhesion through integrins alpha-v beta-3, alpha-v beta-5 and alpha-3 beta-1, so the assay is well matched to studies of stromal remodeling in corneal, renal and hepatic fibrosis models, as well as to oncology work on cell-ECM crosstalk. Sample compatibility covers cell culture supernatants and tissue homogenates in addition to serum and plasma; for sensitive matrices, dilute samples at least 1:2 in assay diluent and avoid more than two freeze-thaw cycles. Each kit supports 96 determinations in strip-well format, with all reagents supplied ready to use except for the wash buffer concentrate and standard reconstitution, and remaining components remain stable at 2-8 degrees C through the printed expiration. Assay Genie provides a lot-specific certificate of analysis and a technical datasheet containing sensitivity, dynamic range and cross-reactivity data so that investigators can adapt the protocol to their own biomarker discovery workflows.
