Human Testoterone ELISA Kit

Assay Genie provides a competitive ELISA kit for the quantitative measurement of testosterone in human biological samples. The colorimetric, antigen-coated competitive ELISA format is intended for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. With a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL, the assay delivers reproducible quantification of this key hormone involved in muscle growth, bone density, and reproductive health.

Performance validation demonstrates consistent linearity and recovery across common sample matrices. Linearity in serum (n=10) ranges from 89-100% at 1:2, 87-99% at 1:4, and 80-97% at 1:8 dilution. EDTA plasma (n=10) gives 87-101%, 87-100%, and 81-98% across the same dilutions, while heparin plasma (n=10) gives 87-102%, 83-95%, and 80-95%. Recovery testing returns 86-105% (average 97%) for serum (n=10), 94-102% (average 98%) for EDTA plasma (n=10), and 88-105% (average 95%) for heparin plasma (n=10).

The workflow follows a standard competitive ELISA protocol: 50 µL of standard or sample plus 50 µL of biotin-labeled antibody are added per well and incubated at 37°C for 45 minutes, followed by five washes and TMB substrate development in the dark at 37°C for 10-20 minutes. After addition of stop solution the colour changes from blue to yellow, and absorbance is read at 450 nm. Kits should be stored sealed at 2-8°C and not cryopreserved.
