Description

A chemiluminescence immunoassay (CLIA) kit from Assay Genie intended for the in vitro quantitative determination of Human TDP43 (Tar DNA Binding Protein 43KDa). It is reactive with human samples and validated for use with serum, plasma and other biological fluids. The procedure uses a sandwich-CLIA design: the microplate wells are pre-coated with an antibody specific for TDP43, after which standards or samples, a biotinylated detection antibody and an Avidin-Horseradish Peroxidase (HRP) conjugate are added sequentially. Unbound components are washed away and the chemiluminescent reaction is read as relative light units (RLU), which are positively correlated with TDP43 concentration and quantified against a standard curve.

According to the manufacturer, the assay runs in 4.5h with a sample volume of 100 μL. Reported sensitivity is 37.50 pg/mL and the detection range is 62.50–4000 pg/mL. The kit is stated to recognise Human TDP43 with no significant cross-reactivity or interference observed with analogues, and reproducibility is reported as both intra-assay and inter-assay CV below 15%. The detection method is chemiluminescence.

Supplied items include a CLIA plate, reference standard, concentrated biotinylated detection antibody (100×), concentrated HRP conjugate (100×), reference standard and sample diluent, biotinylated detection Ab diluent, HRP conjugate diluent, concentrated wash buffer (25×), substrate reagents A and B, desiccant, plate sealer and a product description sheet. Different components require different storage, with the plate, standard, conjugated antibodies and substrate reagents stored at -20 °C and certain diluents kept at 4 °C, all as specified by the manufacturer.
