Transcription Factor 7 (TCF7), also called TCF1, is a high mobility group (HMG) box transcription factor and a downstream effector of the canonical Wnt/β-catenin signaling pathway. TCF7 governs critical steps in T cell lineage commitment within the thymus, including the transition from CD4⁻CD8⁻ double-negative to CD4⁺CD8⁺ double-positive thymocytes. It also drives the long-term self-renewal of memory CD8⁺ T cell populations after infection or vaccination, and its loss is associated with the exhausted T cell phenotype observed in chronic viral infections and tumor microenvironments. In oncology, recurrent TCF7 alterations and altered Wnt pathway activity have been documented in T-cell acute lymphoblastic leukemia and other lymphoid malignancies.

Investigators use this kit for in vitro research, including profiling Wnt pathway activation in primary lymphocytes, characterizing transcription factor expression across T helper and cytotoxic T cell differentiation states, and screening transcriptional signatures in tumor biopsy homogenates and peripheral blood mononuclear cell lysates.
