Human TBPL1/TBP like protein 1 ELISA Kit

Assay Genie provides this Sandwich ELISA, Double Antibody kit for the quantitative detection of human TBPL1 (TATA box-binding protein-like protein 1, Uniprot P62380). The assay is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Performance specifications include a sensitivity of 46.875 pg/mL and a working range of 78.125-5000 pg/mL. The pre-coated capture antibody is paired with a biotin-conjugated detection antibody and HRP-Streptavidin reporting, with absorbance read at 450 nm after TMB substrate development. Reagents are stored at 2-8°C for 12 months.

Sample preparation guidance covers serum, plasma (with EDTA-Na2/K2 recommended as the anticoagulant), cell culture supernatant, cell lysates and tissue homogenates. The microplate workflow proceeds through standard or sample incubation at 37°C for 90 minutes, biotin-labelled antibody binding for 60 minutes, HRP-Streptavidin binding for 30 minutes, and TMB colour development for 10-20 minutes, followed by stop solution and immediate absorbance reading at 450 nm. Duplicate wells are recommended for standards and samples.

Reported linearity in serum (n=5) is 91-104% at 1:2, 84-99% at 1:4 and 81-100% at 1:8; in EDTA plasma (n=5) 87-102%, 87-100% and 80-93%; and in heparin plasma (n=5) 88-105%, 87-101% and 81-92%. Spike-and-recovery performance averages 95% in serum, 99% in EDTA plasma and 97% in heparin plasma, with corresponding recovery ranges of 86-101%, 85-105% and 89-103%.
