Description

A sandwich ELISA from Assay Genie for the quantitative determination of TAR DNA-Binding Protein 43kDa (TDP43) in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a double-antibody sandwich format with colorimetric detection and offers a sensitivity of 0.188 ng/mL across a working range of 0.313–20 ng/mL. TDP43 is a key protein involved in RNA processing and gene regulation, with dysregulation linked to neurodegenerative conditions such as amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration (FTLD).

The protocol follows a standard sandwich workflow: equilibrate reagents, set standards, samples and controls on the pre-coated plate, incubate at 37°C for 90 minutes, add biotin-labelled detection antibody and incubate for 60 minutes, add HRP-streptavidin (SABC) and incubate for 30 minutes, develop with TMB substrate in the dark for 10–20 minutes, then add stop solution and read absorbance at 450 nm. The kit is supplied with a pre-coated dismountable microplate, lyophilized standard, biotin-labelled antibody, HRP-streptavidin conjugate, TMB substrate, sample, antibody and SABC dilution buffers, stop solution, wash buffer and plate sealer, all stored at 2–8°C for 12 months.

Assay performance has been characterised by recovery and linearity testing. Spiked-sample recovery ranged from 89–101% in serum (average 93%), 86–104% in EDTA plasma (average 95%), and 86–103% in heparin plasma (average 95%), with serial-dilution linearity of 80–105% across the same matrices. The Uniprot entry is Q13148, and the protein is also known by the synonyms TDP43, ALS10 and TARDBP.
