Description

From Assay Genie, this colorimetric Sandwich ELISA uses a double-antibody format for the quantitative measurement of TACR3, also known as the Neuromedin K receptor, in human samples. The kit is designed for accurate quantification of TACR3 levels in serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples, making it a useful tool for research in neuroscience, endocrinology, and drug development. TACR3 is a G protein-coupled receptor that mediates the effects of neuromedin K, a neuropeptide involved in regulating pain perception, stress response, and appetite control, and abnormalities in its signalling have been implicated in conditions including schizophrenia, depression, and irritable bowel syndrome, making it a promising target for therapeutic intervention.

Performance characteristics reported by the manufacturer include a detection sensitivity of 0.469 ng/mL and an assay range of 0.781–50 ng/mL. Linearity in serum (n=5) ranged from 85–104% at 1:2, 92–97% at 1:4, and 89–101% at 1:8 dilution; EDTA plasma (n=5) gave 92–97%, 90–101%, and 83–100% at the same dilutions; heparin plasma (n=5) gave 83–96%, 83–94%, and 84–100%. Recovery averaged 93% for serum (range 91–100%), 91% for EDTA plasma (range 87–100%), and 96% for heparin plasma (range 90–101%).

The procedure involves preparing reagents and loading standards, samples and controls onto the pre-coated plate, incubating at 37°C for 90 minutes, adding biotin-labelled detection antibody for a 60-minute incubation, then HRP-streptavidin conjugate for 30 minutes, developing with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm after adding stop solution. Serum is prepared by allowing blood to clot followed by centrifugation at 1000 × g for 20 minutes; plasma is collected using anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2–8°C. Kits should be stored at 2–8°C and remain stable for 12 months under these conditions.
