Description

This Sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of human Tachykinin-3 (TAC3) levels using a double-antibody method with colorimetric detection. Tachykinin 3 is a neuropeptide that plays a vital role in the regulation of various physiological processes, including gastrointestinal functions, pain perception, and inflammation. Dysregulation of Tachykinin 3 has been linked to conditions such as irritable bowel syndrome, asthma, and neurogenic inflammation, highlighting its importance as a potential biomarker for studying these conditions.

The assay reports a sensitivity of 7.5 pg/mL and a working range of 12.5-800 pg/mL. Acceptable sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is stored at 2-8°C for 12 months. The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks, incubating the plate at 37°C for 90 minutes for antigen binding, adding biotin-labeled detection antibody with a 37°C incubation for 60 minutes, binding HRP-Streptavidin (SABC) for 30 minutes, developing colour with TMB substrate in the dark for 10-20 minutes, then adding stop solution and measuring absorbance at 450 nm.

Linearity was demonstrated across dilution ratios of 1:2, 1:4 and 1:8 for serum (87-99%, 83-100%, 83-95%), EDTA plasma (85-99%, 80-98%, 81-99%) and heparin plasma (91-104%, 87-103%, 87-104%), with n = 5 per sample type. Recovery ranges were 95-104% (average 98%) for serum, 89-100% (average 91%) for EDTA plasma and 87-103% (average 96%) for heparin plasma. Sample synonyms listed for the target include NKB, HH10, NKNB, PRO1155, ZNEUROK1, Neurokinin B protein and Neurokinin beta, and the kit is associated with Cell Biology research applications.
