Human T4 ELISA Kit

From Assay Genie, this competitive ELISA kit is designed for the quantitative measurement of human T4 (Thyroxine, also known as Free Thyroxine, fT4, or total T4) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a colorimetric detection method in a Competitive ELISA, Coated with Antigen format and is intended for use with human samples only.

According to the manufacturer, the kit has a sensitivity of 0.938 pg/mL and a measurement range of 1.563–100 pg/mL. Reported linearity at 1:2, 1:4, and 1:8 dilutions (n=10) covers 89–99%, 82–100%, and 84–100% for serum; 89–99%, 84–100%, and 81–97% for EDTA plasma; and 86–100%, 83–94%, and 87–96% for heparin plasma. Recovery values are 92–100% (average 94%) for serum, 92–101% (average 97%) for EDTA plasma, and 88–105% (average 98%) for heparin plasma.

The procedure involves equilibrating TMB substrate for 30 minutes at room temperature, adding 50 µL of standard or sample followed by 50 µL of biotin-labeled antibody per well, incubating at 37°C for 45 minutes, washing five times with a 1 minute soak per wash, developing color at 37°C for 10–20 minutes in the dark, stopping the reaction (blue to yellow), and reading absorbance at 450 nm. Storage is recommended at 2–8°C (sealed); cryopreservation should be avoided. The kit supports research into thyroid function, where Thyroxine (T4), a key hormone produced by the thyroid gland, regulates metabolism, growth, and energy levels, with imbalances linked to thyroid disorders.
