A quantitative sandwich double-antibody ELISA from Assay Genie for measuring endogenous human Syntaxin-12 (STX12), a SNARE-family membrane fusion protein that localises to endosomes and supports vesicle docking, autophagosome maturation, and late endosome-to-lysosome trafficking. The microplate is pre-coated with a capture antibody specific to STX12, and bound analyte is detected with a biotinylated detection antibody followed by HRP-conjugated streptavidin and TMB substrate, giving a colorimetric signal read at 450 nm on a standard plate reader.

It is validated for use with serum, plasma, cell culture supernatant, and cell or tissue lysate, with serum and plasma samples typically prepared by centrifugation at 1000 x g. The antibody pair specifically recognises STX12, with no obvious cross-reaction with analogous SNARE-family proteins, supporting human-specific quantification rather than cross-species work.

Each pack is configured for one 96-well plate and contains a dismountable pre-coated microplate, lyophilised recombinant standard, biotin-labelled detection antibody (100X), HRP-streptavidin concentrate (100X), TMB substrate, sample and antibody dilution buffers, 25X wash concentrate, and stop solution. The workflow pairs a 90 minute primary incubation at 37 °C with a 60 minute detection antibody step and a 30 minute HRP step, completing in roughly four hours excluding equilibration and sample preparation. Unused strips and standards should be returned to a sealed foil pouch with desiccant, where the kit remains stable for 12 months at 2-8 °C; the biotin antibody, SABC, and TMB components should be protected from light and not frozen. Provided for research use only and not validated for diagnostic or clinical decision-making.
