Syncoilin is a type III intermediate filament protein that scaffolds the dystrophin-associated glycoprotein complex to the desmin intermediate filament network in skeletal and cardiac muscle. The Assay Genie Human Syncoilin (SYNC) ELISA Kit is an in vitro immunoassay tool developed for quantitative measurement of SYNC protein in human tissue homogenates, cultured cell lysates, and other research samples. SYNC expression is altered in Duchenne muscular dystrophy and desmin-related myopathies, making this kit valuable for neuromuscular and muscle biology research programs investigating disease mechanisms and potential therapeutic interventions.

Each kit supplies pre-coated microplate wells, biotinylated detection antibody, streptavidin-HRP conjugate, recombinant protein standards for generating a standard curve, wash buffer, substrate, and stop solution. Reagents should remain refrigerated between uses, and the plate should be stored in its sealed foil pouch with desiccant to protect from moisture before opening. Reconstituted standards and antibody working solutions should be aliquoted to limit repeated freeze-thaw cycles, and all components should be equilibrated to room temperature prior to running the assay.

Researchers apply this kit to compare SYNC abundance between healthy and dystrophic tissue samples, evaluate SYNC protein changes following gene therapy or pharmacological treatment in cell or animal models, or profile SYNC expression in patient-derived myogenic cell lines. Standards are diluted in serial fashion to generate a multi-point standard curve, against which unknown sample optical density values are interpolated to determine SYNC concentration. The procedure is supplied for research use only and is not intended for clinical diagnostic applications.
