Description

Manufactured by Assay Genie, this Sandwich ELISA kit enables the quantitative detection of synaptophysin in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Synaptophysin is a key protein found in synaptic vesicles and is essential for neurotransmitter release and synaptic function. The product is positioned for researchers studying neurodegenerative disorders, synaptic plasticity, and neurological diseases, with documented relevance to synaptic transmission and related neurological conditions.

The double-antibody sandwich ELISA uses a colorimetric detection method. Sensitivity is quoted as 0.188 ng/mL and the working range is 0.313–20 ng/mL. Reported linearity for serum dilutions at 1:2, 1:4 and 1:8 (n=5) is 89–105%, 83–101% and 82–98%; EDTA plasma shows 89–104%, 85–97% and 80–95%, and heparin plasma shows 87–105%, 89–101% and 84–99%. Recovery ranges are 87–96% for serum (average 93%), 88–98% for EDTA plasma (average 94%) and 85–99% for heparin plasma (average 93%). The kit should be stored at 2–8°C for 12 months. The associated UniProt entry is P08247; listed synonyms include SYP, Major synaptic vesicle protein p38, MRX, MRXSYP and synaptophysin. Research area is cell biology.

The assay protocol proceeds in six stages: equilibrate reagents and TMB substrate to room temperature and set up standard, sample and control (zero) wells; primary incubation at 37°C for 90 minutes; addition of biotin-labelled detection antibody followed by incubation at 37°C for 60 minutes; HRP-Streptavidin (SABC) binding at 37°C for 30 minutes; colour development with TMB substrate in the dark for 10–20 minutes; and stop reaction with absorbance measurement at 450 nm. Sample preparation guidance covers serum (clot, centrifuge at 1000 × g for 20 minutes), plasma (anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2–8°C), tissue homogenates, cell culture supernatant (centrifuge at 2500 rpm for 5 minutes) and cell lysates. The supplied protocol is sample-based and protocols are specific to each batch/lot.
