Matriptase, the type II transmembrane serine protease also known as Suppression of Tumorigenicity 14 (ST14) and membrane-type serine protease 1 (MT-SP1), can be quantified in human specimens using this immunoassay kit from Assay Genie. ST14 has been a focal point of research into epithelial barrier integrity, keratinocyte terminal differentiation, and pericellular proteolysis, with altered expression patterns reported in carcinoma cell lines and primary tumour specimens. The kit provides coated plate wells, matched detection antibodies, standard calibrators, substrate, and stop solution sufficient for completing a full microplate run of unknown samples alongside an appropriately diluted standard curve.

Validated for in vitro research applications, the assay accepts a broad range of human-derived inputs including serum, EDTA- or heparin-treated plasma, tissue homogenates prepared in non-denaturing buffer, cell lysates, and clarified cell culture supernatants. Read-out is absorbance-based and can be performed on any standard microplate reader. Reagents are supplied ready-to-use or as concentrated stocks, and unopened components should be kept refrigerated and protected from light to preserve activity through the stated shelf life; reconstituted standards should be aliquoted to limit repeated freeze-thaw cycles.

The accompanying protocol covers plate setup, standard curve fitting (typically four-parameter logistic regression), recommended sample dilution ranges, washing parameters, and troubleshooting guidance. Because the antibody pair is directed at the human ST14 ectodomain, results are not directly interchangeable with murine matriptase measurements, and species cross-reactivity has not been demonstrated for this particular configuration. Laboratories investigating tumour invasion, skin homeostasis, or mucosal inflammation pathways will find this reagent useful for tracking shed and secreted ST14 in conditioned media and processed tissue extracts.
