The human stearoyl-CoA desaturase (SCD) enzyme occupies a central position in lipid biosynthesis, catalyzing the introduction of a cis-delta-9 double bond into saturated fatty acyl-CoA substrates and producing monounsaturated fatty acids that fuel membrane synthesis and energy storage. Investigators studying obesity, type 2 diabetes, non-alcoholic fatty liver disease, and tumor lipid metabolism routinely quantify SCD protein abundance as a marker of de novo lipogenesis activity in hepatocytes, adipocytes, and cancer cell lines.

This Assay Genie immunoassay enables quantitative measurement of human SCD in tissue homogenates and cell lysates prepared from cultured lines or biopsy material. A typical workflow involves homogenizing tissue in cold lysis buffer supplemented with protease inhibitors, clarifying the lysate by centrifugation, and loading the supernatant onto the precoated microplate. The two-antibody capture-and-detection configuration confers specificity for the human SCD isoform, with absorbance subsequently read on a standard plate reader for relative quantitation against a calibrator curve.

Each kit supplies reagents sufficient for 96 individual determinations, supporting medium-throughput studies of metabolic pathway regulation, siRNA or CRISPR knockdown validation, and pharmacological screening of candidate lipogenesis inhibitors. Provided in a microplate format, the product is intended for research use only and finds application across adipocyte differentiation models, hepatic steatosis investigations, and studies of diet-induced obesity in rodent and human tissue samples.
