Human STAT5b ELISA Kit

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of STAT5B in human samples using colorimetric TMB substrate development with absorbance read at 450 nm. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay has a stated sensitivity of 0.469 ng/mL and a measuring range of 0.781-50 ng/mL, with unopened components stored at 2-8°C for 12 months.

STAT5B is described as a key transcription factor involved in cell proliferation, differentiation, immune response, cell growth and survival, with dysregulation linked to cancer, autoimmune disorders and inflammatory conditions. Linearity (n=5) at 1:2, 1:4 and 1:8 dilutions was 82-100%, 84-99% and 84-98% for serum, 88-100%, 83-94% and 86-100% for EDTA plasma, and 86-97%, 85-100% and 86-101% for heparin plasma. Mean recoveries were 98% for serum, 97% for EDTA plasma and 98% for heparin plasma.

The workflow equilibrates reagents to room temperature, incubates the loaded plate at 37°C for 90 minutes, adds biotin-labelled detection antibody (37°C, 60 minutes), then HRP-streptavidin for 30 minutes at 37°C, develops colour with TMB in the dark for 10–20 minutes, and stops the reaction before absorbance is read at 450 nm. Supplied components include a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody (100X), HRP-streptavidin conjugate (100X), TMB substrate, sample, antibody and SABC dilution buffers, stop solution, 25X wash buffer, plate sealer and a technical manual.
