Wild-type SS18 partners with BRG1 and BRG5 within the SWI/SNF complex to evict Polycomb repressive complexes and activate transcription; the same locus is pathologically rearranged into SS18-SSX1, SS18-SSX2, and SS18-SSX4 fusion oncogenes that drive synovial sarcoma. Measuring native SS18 levels supports research into tumor biology, translocation mechanism, and chromatin state.

The capture antibody is specific to human SS18, with no significant cross-reactivity with SSX fusion proteins or other SWI/SNF subunits, making the readout useful for distinguishing wild-type expression from translocation-driven contexts. Researchers should aliquot the standard and detection antibody after first thaw to minimize freeze-thaw cycles, refrigerate the sealed plate and reagents at the temperature indicated on the kit insert, and bring all components to room temperature before beginning the assay. The protocol typically uses 50-100 uL of input material and produces a chromogenic end-point read on a standard microplate reader for in vitro research applications.
