The assay uses a competitive ELISA format with antigen-coated wells and delivers a detection sensitivity of 0.188 ng/mL across a working range of 0.312-20 ng/mL. SRY, also known as testis-determining factor (TDF), is a transcription factor central to male sex determination, and abnormalities in the gene are linked to disorders of sex development.

Validated linearity in serum spans 90-103% at 1:2, 89-99% at 1:4, and 88-98% at 1:8 dilution. EDTA plasma shows 88-103%, 88-99%, and 86-99%, while heparin plasma shows 91-101%, 89-101%, and 85-100% across the same dilutions. Recovery testing returned averages of 98% for serum, 94% for EDTA plasma, and 96% for heparin plasma. Kit components should be stored at 2-8°C, protected from direct light and never cryopreserved.
