Human SRC1/RIP160/NCOA1 ELISA Kit

Assay Genie's Human SRC1/RIP160/NCOA1 ELISA Kit is a sandwich (double antibody) immunoassay for the quantitative measurement of the transcriptional coactivator proteins SRC1, RIP160, and NCOA1 in human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The assay uses an antibody pre-coated onto the microplate wells to capture target from standards and samples. After washing, a biotinylated detection antibody is added, followed by HRP-conjugated streptavidin (SABC) and TMB substrate development. The reaction is stopped with an acidic stop solution and absorbance is read at 450 nm. Reagents include a lyophilized standard, biotin-labeled antibody (100X), HRP-streptavidin conjugate (SABC, 100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, wash buffer (25X), plate sealer, technical manual, and Bradford reagent for total protein quantification. Components are stored at 2–8°C unless otherwise indicated.

Validated recovery in serum was 88–103% with an average of 96%; in EDTA plasma 90–104% (average 97%); and in heparin plasma 87–102% (average 94%). Linearity testing at 1:2, 1:4, and 1:8 dilutions gave recovery ranges of 88–104%, 82–97%, and 87–96% for serum; 86–104%, 85–95%, and 80–100% for EDTA plasma; and 85–100%, 88–100%, and 82–91% for heparin plasma.
