The Assay Genie SCCA2 ELISA delivers quantitative measurement of SERPINB4, the squamous cell carcinoma antigen 2 serpin implicated in cervical, lung, esophageal, and head-and-neck tumor biology. Built as a research-use-only tool rather than a clinical diagnostic, it supports biomarker discovery, therapeutic response monitoring, and longitudinal specimen tracking across translational oncology laboratories.

The workflow centres on a pre-coated microplate read against a serpin calibrator dilution series in a 96-assay kit configuration. Each well receives a small microlitre-range sample loading volume before sequential incubation with a biotinylated detection reagent and an enzyme reporter that yields an enzyme-catalysed readout. Accepted matrices span serum fractions, plasma prepared with standard anticoagulants, snap-frozen tissue homogenates, detergent-lysed cell pellets, and clarified cell culture supernatant, giving one protocol the flexibility to interrogate clinical biopsy archives, patient-derived xenograft material, and in vitro cell line experiments within the same laboratory session.

Reactivity characterisation is restricted to the human SERPINB4 antigen, making this kit a deliberate complement to the related SCCA1 (SERPINB3) marker for studies that compute a combined squamous antigen index. That index has been reported in the cervical screening literature as a triage adjunct to cytology and as a candidate biomarker for monitoring treatment response in squamous disease. Components ship as a sealed, cold-chain package suitable for transfer to refrigerated storage upon arrival, with documented component stability supporting longitudinal study planning from the date of receipt.
