Human SPINK1 ELISA Kit

Assay Genie's Human SPINK1 ELISA Kit is a colorimetric Sandwich ELISA (Double Antibody) designed for the quantitative measurement of SPINK1 in human samples. SPINK1, also known as Serine Protease Inhibitor Kazal Type 1, is a protein that regulates protease activity and supports tissue homeostasis, and its dysregulation has been associated with pancreatic cancer, chronic pancreatitis, and inflammatory bowel disease.

Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit delivers a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL, with all reagents stored at 2–8°C and stable for 12 months. The assay workflow involves reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding, HRP-Streptavidin incubation, TMB color development in the dark for 10–20 minutes, and absorbance reading at 450 nm after addition of stop solution.

Linearity has been validated in serum (87–97%, 83–98%, 80–96%), EDTA plasma (88–103%, 87–95%, 81–97%), and heparin plasma (85–101%, 86–94%, 80–99%) across 1:2, 1:4, and 1:8 dilutions. Recovery ranges from 85–99% (serum, average 92%), 90–105% (EDTA plasma, average 96%), and 91–101% (heparin plasma, average 96%). The kit contains a pre-coated dismountable microplate, lyophilized standard, biotin-labeled antibody, HRP-Streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer, plate sealers, and a technical manual.
