SPHKAP, encoded at human chromosomal locus 2q36.3 (HGNC:30619, UniProt Q2M3C7), is a 1671-amino-acid scaffolding protein of roughly 183 kDa that anchors the type I regulatory subunit of cAMP-dependent protein kinase to distinct cytoplasmic microdomains. Its interaction with sphingosine kinase 1 strongly suppresses SPHK1 catalytic activity, positioning SPHKAP as a brake on sphingolipid metabolite production. Tissue distribution is heavily biased toward cardiac muscle, with additional expression in spleen, ovary, and brain regions including amygdala, hippocampus, and thalamus, which is why the Assay Genie ELISA is frequently selected for studies of cardiac remodeling, neuronal lipid signaling, and beta-adrenergic desensitization.
