The colorimetric assay is reactive with human samples and detects both natural and recombinant human S1PR4, supporting investigations into sphingosine 1-phosphate signalling and its role in disease pathogenesis, including inflammatory disorders, autoimmune diseases, and cancer.

Performance specifications include a detection range of 125-8000pg/mL, a sensitivity of 89pg/mL, an intra-assay CV of 5.1%, and an inter-assay CV of 9.5%. Linearity has been validated across dilutions from 1:2 to 1:16 in serum (83-127%), EDTA plasma (95-110%), and heparin plasma (81-117%). The receptor may be involved in cell migration processes specific to lymphocytes and belongs to the G-protein coupled receptor 1 family. As a multi-pass membrane protein located in the cell membrane, S1PR4 is implicated in G-protein coupled receptor protein signalling, including elevation of cytosolic calcium ion concentration and phospholipase C activation.
