Description

Designed for the quantitative measurement of human Sphingosine-1-phosphate phosphatase 1 (SGPP1), this Sandwich ELISA kit from Assay Genie is intended for research use only. The assay is reactive with human samples and is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, with results read colorimetrically following the addition of TMB substrate and stop solution, measuring absorbance at 450 nm.

SGPP1 is an enzyme that specifically dephosphorylates sphingosine-1-phosphate (S1P), dihydro-S1P, and phyto-S1P, and it plays a key part in regulating intracellular S1P levels and the recycling of sphingosine into the sphingolipid synthesis pathway. Dysregulation of SGPP1 has been linked to diseases including cancer, inflammatory disorders, and neurodegenerative conditions, making it a useful biomarker for studying sphingolipid metabolism. The UniProt entry for SGPP1 is Q9BX95, and the enzyme is localised to the endoplasmic reticulum membrane and the cell membrane as a multi-pass membrane protein.

The kit uses a sandwich, double-antibody format and reports a sensitivity of 9.375 pg/mL and a measuring range of 15.625-1000 pg/mL. Linearity (n = 5) is reported across dilutions of 1:2, 1:4, and 1:8 for serum (80-105%), EDTA plasma (83-103%), and heparin plasma (82-105%). Spike-and-recovery performance (n = 5) is given as 91-101% (average 95%) for serum, 85-96% (average 92%) for EDTA plasma, and 88-105% (average 96%) for heparin plasma. The working protocol involves equilibrating reagents to room temperature, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, and HRP-streptavidin binding at 37°C for 30 minutes, followed by colour development in the dark for 10-20 minutes before stopping the reaction.
