Human Sphingomyelin phosphodiesterase/SMPD1 ELISA Kit

Assay Genie offers a sandwich ELISA designed to quantify levels of SMPD1, also known as acid sphingomyelinase, in human biological samples. SMPD1 plays a crucial role in sphingolipid metabolism and contributes to the regulation of cell signaling, apoptosis, and inflammation. Because dysregulation of SMPD1 activity has been linked to conditions such as Niemann-Pick disease and metabolic disorders, accurate measurement of this enzyme is valuable for biomarker and therapeutic research.

This double-antibody sandwich ELISA provides a sensitivity of 0.469 ng/mL and a detection range of 0.781–50 ng/mL. The kit is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents should be stored at 2–8°C, where the kit remains stable for up to 12 months.

The assay follows a straightforward six-step workflow: equilibrate reagents and pre-coat the plate; incubate standards and samples at 37°C for 90 minutes; bind the biotin-labelled detection antibody for 60 minutes; incubate with HRP-Streptavidin for 30 minutes; develop colour with TMB substrate in the dark for 10–20 minutes; and stop the reaction before reading absorbance at 450 nm. Validation data demonstrate linearity and recovery across serum, EDTA plasma, and heparin plasma samples.
