Sperm Equatorial Segment Protein 1 (SPESP1) is a 38 kDa human glycoprotein localized on the equatorial segment of the sperm head, where it participates in membrane interactions between sperm and oocyte during fertilization. The Assay Genie ELISA Kit quantifies native SPESP1 (UniProt Q6UW49) on a pre-coated 96-well strip plate with a 100 µL sample volume, and ships with lyophilized recombinant standard, capture antibody, biotinylated detection antibody, sample diluent, wash buffer concentrate, TMB substrate, and stop solution.

Standards and detection reagents are stored at -20 °C upon arrival while remaining components are kept at 4 °C, supporting a six-month shelf life when freeze-thaw cycles of working aliquots are limited. The assay is validated for human reactivity and is applied by andrology and reproductive biology laboratories to profile SPESP1 abundance in serum, plasma, seminal fluid, testicular tissue homogenates, and cell lysate preparations. Typical workflows include spermatogenesis research, idiopathic fertilization failure investigations, and candidate evaluation for non-hormonal contraceptive targets within translational reproductive medicine programs.
