Human SOST/Sclerostin ELISA Kit

A colorimetric Sandwich ELISA from Assay Genie is designed for the quantitative detection of SOST (Sclerostin) in human samples. The assay is reactive with human samples and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types.

Sclerostin is described as a key regulator of bone metabolism that inhibits bone formation by blocking the Wnt signaling pathway, with altered SOST levels linked to osteoporosis, arthritis, and bone fractures. The kit provides a sensitivity of 37.5 pg/mL and a working range of 62.5–4000 pg/mL, and is intended for research into bone health and related disorders. Reported linearity spans roughly 83–107% across serum, EDTA plasma, and heparin plasma dilutions, with average recoveries between 94–97% in those matrices.

The supplied procedure uses a pre-coated plate: standards and samples are incubated at 37 °C for 90 minutes, followed by a biotinylated detection antibody (60 min), HRP-Streptavidin (30 min), TMB substrate development in the dark, and absorbance readout at 450 nm after stop solution. Kit components include a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody (100×), HRP-Streptavidin conjugate (SABC, 100×), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25× wash buffer, plate sealer, and a technical manual, to be stored at 2–8 °C for up to 12 months.
