Description

From Assay Genie, this colorimetric Sandwich ELISA kit is designed for the quantitative measurement of human sorbitol dehydrogenase (SORD) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The double-antibody sandwich format provides a sensitivity of 0.188 mIU/mL across a measuring range of 0.313–20 mIU/mL. SORD (also known as SDH or L-iditol 2-dehydrogenase; UniProt Q00796) is a key enzyme in sorbitol metabolism, with dysregulation implicated in diabetes and diabetic retinopathy.

Reagents remain stable for 12 months at 2–8°C, and the microplate and lyophilized standard are also suitable for long-term storage at -20°C. Validated linearity spans 85–104% for serum, 85–101% for EDTA plasma, and 84–98% for heparin plasma at a 1:2 dilution, with comparable values through 1:8. Spike-and-recovery averages of 96% (serum), 94% (EDTA plasma), and 98% (heparin plasma) support reliable quantitation across these matrices.

The procedure begins by equilibrating reagents and TMB substrate to room temperature, then loading standards, samples and controls onto the pre-coated plate for a 90-minute incubation at 37°C. A biotin-labeled detection antibody is added for a 60-minute incubation at 37°C, followed by HRP-streptavidin conjugate for 30 minutes at 37°C. TMB substrate is then added for 10–20 minutes in the dark, the reaction is stopped, and absorbance is read at 450 nm. The kit is intended for cell biology research investigating SORD biology and its role in disease.
