The human sodium iodide symporter (NIS) is a basolateral membrane glycoprotein expressed primarily in thyroid follicular cells, where it actively concentrates iodide from the bloodstream as the first step in thyroid hormone biosynthesis. Encoded by the SLC5A5 gene, NIS serves as the molecular target of radioiodine therapy in differentiated thyroid carcinoma, and loss of functional NIS expression is a well-characterized mechanism behind radioiodide-refractory disease.
