Sodium/Glucose Cotransporter 2 is the renal tubular transporter responsible for the majority of filtered glucose reabsorption, and it sits at the center of investigations into diabetes pathophysiology, glycosuria mechanisms, and the pharmacology of gliflozin-class therapeutics including canagliflozin, dapagliflozin, and empagliflozin.

The assay accepts tissue homogenates and cell lysates as input, fitting studies in renal cortex biopsy material, cultured proximal tubule cell lines, and heterologous expression systems used to model transporter regulation under hyperglycemic or pharmacological challenge. Reagents are supplied refrigerated and should be kept cold between uses to preserve antibody binding activity; allow components to equilibrate to room temperature and aliquot working solutions as needed before initiating the run.

Intended for research use only, the workflow is carried out entirely in vitro and produces a chromogenic read-out whose signal correlates with SGLT2 abundance in the loaded sample.
