The SMCP protein localizes to the outer dense fibers and mitochondrial sheath of the sperm flagellum, where its cysteine-rich regions contribute to structural integrity during capacitation and motility. Mutations and altered expression of SMCP have been linked to asthenozoospermia and idiopathic male infertility, making quantitative measurement valuable for andrology and reproductive biomarker studies.
