Description

A sandwich ELISA from Assay Genie designed for the quantitative measurement of human SIRT7. The assay uses a double-antibody (capture and detection) format with a colorimetric readout at 450 nm following TMB substrate development, and is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate and other liquid samples. Reported performance includes a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL.

Linearity in serum was 89–103% (1:2), 91–103% (1:4) and 87–103% (1:8); in EDTA plasma 82–97%, 87–101% and 84–94%; and in heparin plasma 83–100%, 81–96% and 84–99%. Spike-and-recovery across the same three matrices averaged 98% (serum), 93% (EDTA plasma) and 90% (heparin plasma), with individual ranges of 90–105%, 85–100% and 86–93% respectively. The target corresponds to UniProt Q9NRC8 and is also known as SIRT7, NAD-dependent deacetylase sirtuin-7, SIR2L7, Regulatory protein SIR2 homolog 7 and SIR2-like protein 7.

The supplied protocol follows a sequential incubation scheme: 90 min at 37 °C for antigen binding, 60 min with biotinylated detection antibody, 30 min with HRP–streptavidin (SABC), 10–20 min in the dark for TMB colour development, and absorbance reading at 450 nm after stop solution. Reagents are intended to be stored at 2–8 °C, with the lyophilised standard and plate strips also stable for 12 months at −20 °C. SIRT7 is a member of the sirtuin family involved in gene expression, metabolism and aging, and has been linked to cancer, metabolic disorders and neurodegenerative conditions.
