Human SIRT6 ELISA Kit

A colorimetric sandwich ELISA from Assay Genie designed for the quantitative measurement of human SIRT6 (NAD-dependent deacetylase sirtuin-6, also referred to as SIR2L6 / SIR2-like protein 6; Uniprot Q8N6T7), a protein implicated in maintaining genomic stability, regulating metabolism, influencing aging, and associated with cancer, diabetes and neurodegenerative disorders. The double-antibody sandwich format uses a pre-coated capture antibody and a biotin-conjugated detection antibody, with HRP-Streptavidin and TMB producing a colour change read at 450 nm. It is intended for use with serum, plasma, cell culture supernatant, cell or tissue lysates and other biological fluids, and has a reported sensitivity of 46.875 pg/mL with a working range of 78.125-5000 pg/mL.

Reported linearity in serum (n=5) is 89-105% at 1:2, 85-100% at 1:4 and 84-93% at 1:8; in EDTA plasma (n=5) 86-99%, 82-100% and 81-98%; and in heparin plasma (n=5) 85-103%, 90-99% and 90-100% at the same dilutions. Spike-and-recovery values are 89-103% (average 96%) in serum, 85-103% (average 94%) in EDTA plasma, and 93-104% (average 99%) in heparin plasma. The procedure involves incubating standards and samples for 90 minutes at 37 °C, adding biotin-labelled detection antibody for 60 minutes, HRP-Streptavidin for 30 minutes, and TMB substrate for 10-20 minutes in the dark before stopping and reading at 450 nm. Reagents are shipped on ice packs and stored at 2-8 °C for 12 months.
