Description
This Sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of SIRT4 (NAD-dependent ADP-ribosyltransferase sirtuin-4, also known as SIR2L4 / SIR2-like protein 4) in human samples. The assay uses a double-antibody sandwich format with a colorimetric readout and is reactive with human specimens, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. SIRT4 is a member of the sirtuin family involved in regulating metabolic processes, cell survival and mitochondrial function, and its dysregulation has been implicated in cancer, metabolic disorders and neurodegenerative conditions, making the kit a useful tool for research into these areas.
Spike-and-recovery values across matrices are 86–96% (average 90%) for serum, 87–94% (average 91%) for EDTA plasma, and 91–97% (average 93%) for heparin plasma. Linearity at 1:2, 1:4 and 1:8 dilutions falls within 88–104%, 89–97% and 86–105% for serum; 90–98%, 82–91% and 82–100% for EDTA plasma; and 82–95%, 80–92% and 85–100% for heparin plasma. The protocol involves equilibrating reagents, incubating standards and samples on the pre-coated plate at 37°C, binding a biotin-labelled detection antibody, adding HRP-Streptavidin, developing with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm after adding stop solution. The kit is stored at 2–8°C and is stable for 12 months under those conditions.
