Human SIRP alpha ELISA Kit

Manufactured by Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of SIRP Alpha in human samples. It employs a double antibody sandwich format with colorimetric detection and is suited to research applications in cell biology. SIRP Alpha (also referenced by synonyms including SIRPa, BIT, CD172a, MFR, MYD-1, and SHPS1; UniProt P78324) is a key protein involved in regulating immune responses and cell signalling, and its dysregulation has been linked to cancer, autoimmune disorders, and inflammatory conditions.

Linearity has been validated in serum (85–100% at 1:2; 82–97% at 1:4; 81–100% at 1:8), EDTA plasma (92–103% at 1:2; 80–96% at 1:4; 85–98% at 1:8), and heparin plasma (93–103% at 1:2; 83–94% at 1:4; 83–98% at 1:8). Recovery rates average 92% for serum (range 88–97%), 93% for EDTA plasma (88–99%), and 91% for heparin plasma (82–100%).

The procedure involves equilibrating reagents and loading standards and samples for incubation at 37°C for 90 minutes, adding biotin-labelled detection antibody for 60 minutes, adding HRP-Streptavidin (SABC) for 30 minutes, developing with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit is supplied with an ELISA microplate, lyophilised standard, biotin-labelled antibody, HRP-Streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer, plate sealer, and technical manual, and should be stored at 2–8°C for 12 months.
