Description

Assay Genie offers a high-quality Sandwich ELISA Kit for the quantitative detection of human Signal transducer and activator of transcription 2 (STAT2), also referred to as p113. The assay employs a colorimetric detection method and is validated for use with human samples including serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The kit provides a detection range of 78-5000pg/mL and a sensitivity of 40.1pg/mL, and detects both natural and recombinant human STAT2.

Performance characteristics include an intra-assay CV of 7.8% and an inter-assay CV of 8.9%. Linearity in serum (N=5) ranges from 87-97% at 1:2 dilution to 90-102% at 1:16 dilution, with EDTA plasma values of 99-111% to 107-117% and heparin plasma values of 90-90% to 109-118% across the same dilution series. Recovery in serum averages 106% (range 100-112%) and in plasma 108% (range 102-114%).

STAT2 is a transcription factor of the STAT family that mediates signaling by type I IFNs (IFN-alpha and IFN-beta). Following type I IFN binding to cell surface receptors, JAK kinases (TYK2 and JAK1) are activated, leading to tyrosine phosphorylation of STAT1 and STAT2. The phosphorylated STATs dimerize and associate with IRF9/ISGF3G to form a complex termed ISGF3, which enters the nucleus and binds the IFN stimulated response element (ISRE) to drive an antiviral transcriptional programme. The protein is localised to the cytoplasm and nucleus (UniProt P52630). For research use only.
