Human SHP-2 PharmaGenie ELISA Kit (SBRS1088)

The SHP-2 protein quantified by this Assay Genie kit is a cytoplasmic non-receptor tyrosine phosphatase encoded by the PTPN11 gene. Its two N-terminal SH2 domains keep the catalytic PTP domain autoinhibited under basal conditions, and engagement by phosphotyrosine ligands releases this inhibition, positioning SHP-2 as a positive regulator of the RAS/MAPK cascade and a modulator of PI3K/AKT and JAK/STAT signaling downstream of receptor tyrosine kinases and cytokine receptors.

Research applications include studies of germline gain-of-function mutations that cause Noonan syndrome (PTPN11 variants account for approximately half of affected individuals) and juvenile myelomonocytic leukemia, as well as loss-of-function mutations underlying LEOPARD syndrome. The microplate-based assay is intended for in vitro research use in oncology, developmental biology, and inflammation, including investigations of growth factor responses and downstream effector activation in cell culture systems.
