The Assay Genie quantitative immunoassay enables measurement of human SHARPIN protein, a core component of the linear ubiquitin chain assembly complex (LUBAC) that also contains HOIP and HOIL-1L. SHARPIN contributes a ubiquitin-like domain and an NZF zinc finger to LUBAC, supporting linear (M1-linked) ubiquitination of substrates such as NEMO and RIPK1, which in turn drives canonical NF-κB activation and protects cells from TNF-induced apoptosis.

The Assay Genie kit is configured for 96 Assays and is supplied as a complete reagent set in a colorimetric format intended for research use only. It supports investigations into chronic proliferative dermatitis, inflammatory bowel disease, hepatocellular carcinoma, and B-cell lymphoma, conditions in which altered SHARPIN expression or disrupted linear ubiquitination has been reported in peer-reviewed literature.

SHARPIN was originally identified as SHANK-associated RH domain interactor and independently as SIPL1, and loss-of-function mutations in the corresponding mouse gene produce the chronic proliferative dermatitis (cpdm) phenotype. Quantifying circulating SHARPIN levels can therefore complement genetic and cell-based studies of LUBAC integrity, TNF-driven inflammatory cascades, and the regulation of linear ubiquitin networks in human disease.
