Description

A sandwich ELISA from Assay Genie designed for the quantitative measurement of human Serum paraoxonase/arylesterase 1 (PON1), also referred to as Aromatic esterase 1, K-45, Serum aryldialkylphosphatase 1, A-esterase 1, and PON. The assay is reactive with human samples and accepts serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, using a colorimetric detection method. Reported performance characteristics include a detection range of 1.56-100ng/mL, a sensitivity of 0.78ng/mL, an intra-assay CV of 4.5% and an inter-assay CV of 7.1%.

Linearity was demonstrated in serum, EDTA plasma and heparin plasma at 1:2, 1:4, 1:8 and 1:16 dilutions, with reported ranges of 94-104%, 81-94%, 109-118% and 106-116% for serum (n=5); 109-119%, 105-117%, 87-96% and 107-119% for EDTA plasma (n=5); and 91-101%, 88-100%, 107-116% and 99-110% for heparin plasma (n=5). Spike-and-recovery testing gave average values of 101% (range 95-107%) for serum and 103% (range 97-109%) for plasma. The kit is specific for natural and recombinant human PON1.

PON1 hydrolyses toxic metabolites of a variety of organophosphorus insecticides and is capable of hydrolysing a broad spectrum of organophosphate substrates and lactones, as well as a number of aromatic carboxylic acid esters. The enzyme mediates enzymatic protection of low-density lipoproteins against oxidative modification, a process linked to atheroma formation, and is therefore relevant to research into cardiovascular health, oxidative stress and inflammation. The UniProt reference is P27169 and the kit is supplied for research use only, with the research area listed as Neurosciences.
