Human SERPINB13 ELISA Kit

From Assay Genie, this competitive ELISA kit is designed for the quantitative measurement of human SERPINB13. The assay is coated with antigen and is intended for research in cell biology, where SERPINB13 (also known as Serpin B13, Hurpin, Headpin, PI-13, Peptidase inhibitor 13, Proteinase inhibitor 13, or HaCaT UV-repressible serpin) is a serine protease inhibitor that may play a role in the proliferation or differentiation of keratinocytes.

Validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples of human origin, the kit reports a sensitivity of 0.938 ng/mL and a working range of 1.563–100 ng/mL. Reported linearity at 1:2, 1:4, and 1:8 dilutions spans 81–104% across serum, EDTA plasma, and heparin plasma, with average spike-and-recovery of 96% in serum, 99% in EDTA plasma, and 98% in heparin plasma. Reagents should be stored sealed at 2–8°C and must not be cryopreserved.

The protocol involves equilibrating TMB substrate for 30 minutes at room temperature, adding 50 µL of standard or sample together with 50 µL of biotin-labelled antibody and incubating at 37°C for 45 minutes, washing, developing colour with TMB substrate in the dark at 37°C for 10–20 minutes, terminating with stop solution (a blue-to-yellow colour change), and reading absorbance at 450 nm. The kit includes a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody concentrate, HRP-streptavidin conjugate, TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, wash buffer, plate sealer, and a technical manual.
