Human Serpin H1 (SERPINH1) ELISA Kit

An Assay Genie sandwich ELISA designed for the precise measurement of Serpin H1 (SerpinH1), also known as Hsp47, a heat shock protein that plays a critical role in collagen biosynthesis and quality control. The kit is reactive with human samples and validated for serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, for research use only.

The assay delivers a detection range of 125-8000 pg/mL with a reported sensitivity of 64 pg/mL. Intra-assay CV is 4.7% and inter-assay CV is 8.3%, with specificity for natural and recombinant human Serpin H1.

Dilutional linearity in serum (N=5) spans 94-102% to 94-106% across 1:2 to 1:16 dilutions, with EDTA plasma at 85-95% to 92-102% and heparin plasma at 98-108% to 93-105%. Recovery is 98% (range 92-104%) in serum and 100% (range 94-106%) in plasma. The target protein (UniProt P50454) localises to the endoplasmic reticulum lumen, where it binds specifically to collagen as a chaperone in the biosynthetic pathway of collagen. Abnormal SerpinH1 levels have been linked to fibrosis and cancer, and autoantibodies to the protein have been observed in patients with rheumatoid arthritis.
